Start with a sterile petri dish containing an agar solution. You can make this solution yourself or purchase ready-to-use agar plates. Ensure you open the dishes under strictly sterile conditions to prevent contamination.
Gently scrape a small amount of spores from the print directly onto the surface of the agar in the petri dish. Use a sterile scalpel that has been briefly flame-sterilized for maximum purity.
Seal the petri dish tightly (e.g., with parafilm) and place it in a controlled, dark environment. Here, the spores can germinate and develop into mycelium. This process takes an average of several days to weeks.
Once the mycelium has spread across the agar, look for the most vigorous, white, and clean areas. Use a sterile scalpel to carefully select a small piece of this vital mycelium.
Cut out a piece of healthy mycelium (approx. 6x6 mm) and transfer it to a new agar plate or another suitable growth medium. Continue to work strictly sterile during this procedure.
Repeat the process of isolating and transferring until you have achieved a completely pure, evenly growing culture. Once stable, you can use the mycelium for further cultivation applications.
