Do you already have a ready-to-use liquid culture syringe at home? Then it’s time for the most important step: inoculating your sterilized substrate. What do you need for a sterile inoculation?

Make sure you have all the following materials within reach before you begin:

  • The liquid culture syringe and a new needle: Store the syringe in the refrigerator until right before use.
  • Sterilized substrate: Glass mason jars with grain or grow bags equipped with a (rubber) injection port and a microfilter.
  • 70% Isopropyl Alcohol: For thoroughly disinfecting your workspace.
  • Sterile alcohol wipes: For sanitizing the injection port and any taped areas.
  • Sterile gloves and a face mask: To prevent transferring bacteria from your skin or breath.
  • Blowtorch or jet lighter: Only needed if you are injecting multiple jars in a row, or if you accidentally contaminate the needle (e.g., by touching it).
  • Airtight workbox (Still Air Box): Optional, but highly recommended to block swirling dust particles and drafts.

How much liquid culture do you need?

  • Small glass jar (300 ml): Use approx. 2.0 to 2.5 ml
  • Medium glass jar (500 ml): Use approx. 3.0 to 4.0 ml
  • Large glass jar / substrate (1 kg): Use approx. 6.0 to 8.0 ml
  • Large grow bag (2.5 kg): Use approx. 15.0 to 20.0 ml

Tip for maximum certainty: If in doubt (or always, just to be safe), test your liquid culture on an agar plate first (Agar test). That way, you know for sure that your culture is pure and contaminant-free before injecting it into a large amount of precious substrate.

While injecting more liquid can speed up the colonization process slightly, it also significantly increases the moisture level in your jar or grow bag. Too much moisture quickly leads to mold, bacterial rot (wet spot), and other unpleasant contaminations. When in doubt, always use less.

Theoretically, a tiny drop of liquid culture is enough to eventually colonize an entire substrate; after all, every drop contains countless living mycelium cells. However, using an extremely small amount is not recommended. If you use too little, the colonization process will take significantly longer. The longer large areas of the substrate remain uncolonized, the more opportunity harmful microorganisms have to strike.

Step-by-step: Inoculating your substrate

Follow these four steps carefully for the highest success rate in your grow project.

Reminder: Have you done an agar test yet? It is highly recommended to test your liquid culture on an agar plate first. Are you sure your liquid is pure and uncontaminated? Then proceed directly to the steps below.

Step 1: Disinfecting the workspace and injection port

Close all windows and doors in the room to stop air currents. Generously mist your workspace with 70% isopropyl alcohol and let it sit for a minute. Wash your hands, put on your sterile gloves, and wear your face mask. Then thoroughly wipe down the injection port—or the spot on your jar/bag where you plan to inject—with a sterile alcohol wipe. Finally, gently shake the liquid culture syringe back and forth to distribute the mycelium evenly throughout the liquid.

Step 2: Preparing the needle

Remove the syringe and needle from their original packaging. A brand-new needle straight out of the package is factory-sterilized. Therefore, you do not need to heat it glowing red for the first injection. Twist the needle onto the syringe, remove the protective cap, and you are ready to inject.

Important exception: Did you accidentally touch the exposed needle with your fingers, bump into something, or leave the needle uncapped in the air for a long time? Then the sterility has been compromised. In that case, you must sterilize the needle first by heating it (flame sterilization) and allow it to cool for 10–15 seconds before injecting. Flame sterilization is more effective than using alcohol wipes or 70% isopropyl alcohol. If you truly don't have a flame, alcohol is an option, but significantly less ideal. Inoculating multiple jars in a row? Preferably flame sterilize the needle briefly between injections, or use a new alcohol wipe if no flame is available.

Step 3: Injecting and distributing

Insert the clean needle at an angle through the sterilized injection port of the substrate. Inject a small portion of the liquid along the inside wall of the glass or bag. Without pulling the needle completely out, gently angle the syringe in another direction and inject a little more liquid. By distributing the liquid in three or four different directions, you create multiple starting points for growth.

Step 4: Sealing and incubation

Pull the needle out of the injection port in one smooth motion. If you're using a self-healing port, the hole seals automatically and you don't need to do anything further.

Does your grow bag or jar lack a self-healing injection port? Crucially, wait until the alcohol has completely evaporated before sticking the included patch/sticker over the hole. If the alcohol hasn't fully evaporated, the adhesive layer won't stick and the sticker will peel off, dramatically increasing the risk of contamination. If you don't have a special filter sticker, a piece of micropore tape works great as an alternative.

Finally, place your inoculated and well-sealed substrate in a clean, dark place at a constant room temperature of 21°C to 23°C.

Common mistakes & important points of attention

  • Forgetting to shake beforehand: The mycelium inside the syringe can clump together in the fridge. Gently shake the syringe back and forth right before use to evenly distribute the mycelium in the liquid.
  • Overdosing liquid: Injecting too much liquid culture is a classic beginner's mistake. It displaces oxygen at the bottom of the jar, drastically increases humidity levels, and causes mold or "wet spot" (anaerobic bacterial rot).
  • Not letting the needle cool down (when flame sterilizing between uses): Did you decide to flame sterilize the needle (e.g., after touching it or between two jars)? Always let it cool for 10 to 15 seconds. If you inject immediately, the hot needle will cook and kill the living mycelium.
  • Skipping the agar test when in doubt: Injecting an untested or long-stored culture directly into a large bag of grain can lead to wasting your substrate if there happens to be a hidden contamination in the liquid.
  • Placing substrate directly on a heat mat: Never put grow jars or bags directly on a heat source. Direct heat evaporates moisture at the bottom of the container, drying out the culture underneath and drastically increasing the risk of contamination. Always ensure an insulating layer of air or a gap of at least 2 to 3 centimeters.
  • Working in a draft: Swirling dust and air currents move invisible mold spores. Ensure the room is closed off, or work in a Still Air Box for the highest chance of success.

Frequently Asked Questions

Why should I do an agar test before inoculating?
Although a ready-to-use liquid culture is theoretically sterile, an agar test gives you 100% certainty. Especially if your syringe is a bit older or if you doubt its purity, this is recommended. By applying one or two drops to an agar plate, you can see within a few days whether pure, white mycelium is growing. Do you see strange colors or cloudiness? Then the liquid is contaminated. An agar test prevents you from needlessly throwing away a large amount of valuable substrate.
How long can I store a liquid culture syringe?
An unused liquid culture syringe is best stored in the refrigerator at a temperature between 2°C and 8°C. The mycelium will go into a dormant state, keeping the syringe viable for about 6 to 12 months. Make sure it doesn't freeze, as that destroys the cells. Storing the syringe at room temperature? Then viability drops quickly, and shelf life is a maximum of 1 to 2 months.
What happens if I inject too much liquid culture?
Too much liquid in your jar or grow bag disrupts the moisture balance. The excess moisture sinks to the bottom and displaces oxygen there. This creates the ideal environment for anaerobic bacteria, leading to 'wet spot' (bacterial rot). Your substrate will start smelling sour, become slimy, and mycelium will stop growing. Therefore, always strictly adhere to the recommended dosage.
When will I see mycelium growing for the first time?
A major advantage of liquid culture over a spore syringe is speed. Because the mycelium is already living and active, it no longer needs to germinate. You will usually see the first white, fluffy, or rope-like strands (rhizomorphic growth) appear within 3 to 7 days after injection. Depending on temperature and substrate volume, full colonization is completed within 1 to 3 weeks.
Should I place my grow bag or jar on a heat mat after injecting?
No, never place your substrate directly on a heat mat! Direct contact causes heat to seep into the bottom of the container. This evaporates moisture at the bottom, drying out the culture underneath while creating condensation at the top, which dramatically increases the risk of contamination. Always ensure a stable room temperature (21°C - 23°C). Do you still need to use a mat because the room is very cold? Make sure there is an insulating layer of air or a gap of at least 2 to 3 centimeters between the mat and the grow container.
